Sijunzi decoction on gastric MUC5AC expression of spleen deficiency rats

摘要: [目的]探讨脾虚证大鼠胃黏膜黏液凝胶(MUC5AC)的变化情况,以及健脾益气中药四君子汤对其改变的影响。[方法]除空白对照组大鼠外,其余3组均采用复合因素造模法复制脾虚证动物模型。脾虚证模型组造模7周;自然复健组、四君子汤组造模4周后常规喂养3周,四君子汤组同时用四君子汤进行治疗。用Western bolt方法检测观察各组大鼠胃组织中MUC5AC表达的变化。[结果]与空白对照组比较,脾虚证模型组、自然复健组大鼠胃组织中MUC5AC含量明显减少(P<0.05),四君子汤组的MUC5AC含量明显增加(P<0.05)。[结论]脾虚证可以通过对胃黏膜的防御机制的微观指标产生影响,从而导致胃黏膜屏障功能的下降,进一步导致慢性胃病的发生;四君子汤可通过改善胃黏膜屏障功能,从而对慢性胃病起到治疗作用。Abstract: [Objective] To investigate the gastric MUC5AC expression in spleen deficiency rats,and to detect the effect of Sijunzi decoction (SD) on the expression.[Methods] Composite factors were used to duplicate the animal model of spleen deficiency and SD was applied for the treatment.MUC5AC expression was detected by western bloting in rat stomach tissue.[Results] MUC5AC expression was decreased significantly in spleen deficient models (P<0.05),and after given SD treatment,MUC5AC expression increased significantly (P<0.05).[Conclusion] Spleen deficiency has micro-changes on the gastric mucosa which lead to its decreased barrier function. SD, a kind of herbal composition with effect of strengthening spleen Qi, can improve the micro-changes such as MUC5AC expression and treat the chronic gastric diseases.
