Effect of bilirubin on the cell proliferation and cell cycle in hepatic stellate cell

摘要: [目的]观察胆红素对肝星状细胞(HSC)-T6增殖及细胞周期影响。[方法]将培养细胞分成正常组和胆红素不同浓度(10 μmol/L、30 μmol/L、50 μmol/L、70 μmol/L、100 μmol/L)干预组,采用MTT法观察胆红素对HSC-T6增殖的影响,流式细胞仪观察各组细胞周期的变化。[结果]①不同浓度胆红素对HSC-T6均有促进增殖作用,且呈一定的量效关系,与正常组比较差异有统计学意义(P<0.05);②10 μmol/L、50 μmol/L、100 μmol/L浓度胆红素作用HSC-T6后,G0/G1期减少,S期增加,G2/M期增加,与正常组比较均P<0.05。[结论]胆红素对HSC-T6均有促进增殖作用。Abstract: [Objective] To explore the effect of bilirubin on the cell proliferation and cell cycle in hepatic stellate cell(HSC)-T6.[Methods] The immortalized rat hepatic stellate cell line HSC-T6 was commonly cultured after revival,and was allocated into normal control group and treatment groups with different concentrations bilirubin.The cell proliferation was assessed by using MTT assay,and cell cycle was analyzed by flow cytometry.[Results] ①After intervention of different concentration bilirubin(the extreme concentration was 10 μmol/L,30 μmol/L,50 μmol/L,70 μmol/L,100 μmol/L respectively),the A value increased significantly(compared with the control group,all P<0.05)and showed a dose-dependent manner.②In cell cycle progression,the cell at G0/G1 phase decreased obviously but increased at S and G2/M phase when bilirubin concentration at 10 μmol/L,50 μmol/L,100 μmol/L(compared with the control group,P<0.05).[Conclusion] Bilirubin can promote the proliferation of HSC-T6.
