Effects of DOK3 gene expression on the proliferation,invasion and migration ability of HGC-27 gastric cancer cell

摘要: [目的]分析中晚期胃癌患者DOK3表达情况及对HGC-27胃癌细胞增殖与侵袭迁移能力产生的影响。[方法]选择120例2015年12月~2017年7月收治的胃部疾病患者,以其疾病类型差异分组,A组为低级别上皮内瘤变患者(20例),B组为高级别上皮内瘤变患者(40例),C组为慢性浅表性胃炎患者(40例),D组为中晚期胃癌患者(20例)。对其DOK3表达展开专业检测,并记录其HGC-27细胞组织增殖状况及其侵袭迁移状况。[结果]A组DOK3表达阳性率为60.00%,B组为50.00%,C组为82.50%,D组为35.00%,B组及D组比C组低,差异有统计学意义(P<0.05);C组与A组对比、A组与B组对比差异无统计学意义(P>0.05);对HGC-27细胞组织中DOK3基因展开敲降后,对于胃癌细胞组织的增殖产生促进作用,且细胞组织侵袭迁移作用会更为明显。[结论]胃部癌变现象发生的生物学指标之一即DOK3,且DOK3同时还是抑癌基因类型,在中晚期胃癌患者临床诊断中具有利用价值。Abstract: [Objective]Analysis the DOK3 expression on the advanced gastric cancer patients and its influence on the proliferation of HGC-27 gastric cancer cells and invasive migration capability.[Methods]One hundred and twenty cases of gastric diseases patients was extracted from December 2015 to July 2017,randomly divided into control group and experimental group according to differences in disease types,group A has 20 cases of low grade intraepithelial neoplasia patients,group B has 40 cases of high-grade intraepithelial neoplasia patients,group C has 40 cases of chronic superficial gastritis patients,group C has 20 cases of advanced gastric cancer patients,all of these patients were detection of DOK3 expression,and record the proliferation and invasion and migration of HGC-27 cells.[Results]The positive rate of DOK3 expression in group A was 60.00%,group B was 50.00%,group C was 82.50%,and group D was 35.00%,group B and group D were lower than those of the group C,with statistically significant(P<0.05),there were no significant difference between group C and A,group A and B(P>0.05).After knock down the DOK3 gene in the cell organization of HGC-27,it can promote the proliferation of gastric cancer cell tissue,and the invasion and migration of cells and tissues will be more obvious.[Conclusion]One of the biological indicators of gastric carcinogenesis is DOK3,and DOK3 is also a tumor suppressor gene type,it has value in clinical diagnosis of patients with advanced gastric cancer.
